Following her debut as a child in the 1977 film Karm, Matondkar rose to prominence with Masoom (1983), after which she appeared in a few other films. Her first adult role in Malayalam cinema was in Chanakyan (1989), and her first adult role in Hindi cinema was in Narsimha (1991), both of which were commercial successes. Matondkar, in addition to acting in films, is involved in a number of humanitarian causes and is outspoken about issues affecting women and children. She has performed in concert tours and stage shows, as well as served as a talent judge on various dance reality shows such as Jhalak Dikhhla Jaa.
